AlloDx SuperNAT: Advancing Nephrin Antibody Testing in Podocyte Disease

In recent years, Nephrin antibodies have attracted increasing attention from the nephrology community as potential biomarkers for podocyte-related diseases such as minimal change disease (MCD) and focal segmental glomerulosclerosis (FSGS). SuperNAT second-generation highly sensitive Nephrin antibody detection technology independently developed by AlloDx has achieved remarkable technological breakthroughs.


In recent years, Nephrin antibodies have attracted increasing attention from the nephrology community as potential biomarkers for podocyte-related diseases such as minimal change disease (MCD) and focal segmental glomerulosclerosis (FSGS). The SuperNAT second-generation highly sensitive Nephrin antibody detection technology independently developed by AlloDx was invited to conduct a poster presentation at ASN Kidney Week 2025 (DOI: 10.1681/ASN.2025j525v94y) for its significant technological breakthroughs and related research results, showing good academic and clinical value.

01 Dual innovation strategy to improve detection capabilities

In order to improve the accuracy of Nephrin antibody detection, AlloDx systematically optimized the traditional ELISA method and developed the Super Nephrin Antibody Trap (SuperNAT) detection technology. This technology uses a dual innovative strategy of magnetic bead antigen capture combined with background subtraction, which can effectively overcome the interference of high-abundance serum proteins such as rheumatoid factor and heterophilic antibodies, significantly improve the detection sensitivity of extremely low-abundance Nephrin autoantibodies, with a minimum detection limit of 3 U/mL, and still has excellent anti-interference performance in complex sample matrices.

In clinical verification, SuperNAT demonstrated excellent detection capabilities. In 40 patient samples, the antibody level detected by this method was significantly higher than that of the healthy control group (59.71 vs. 9.22 U/mL, p < 0.0001); when 45 U/mL was used as the positive determination threshold, the positive detection rate reached 55%, which was 48.6% more sensitive than the traditional ELISA method. In addition, the research team also successfully identified two potential immune epitopes of Nephrin protein (15 aa–29 aa and 778 aa–793 aa), providing new clues to reveal the immune mechanism of related diseases.

This technology also has good sample stability (quantitative difference within 1 month ≤7%) and excellent automation compatibility, laying the foundation for large-sample clinical screening and routine testing.

02 Nephrin kit has been patented

Currently, AlloDx has applied for a patent (202511390182.2) for the Nephrin antibody detection kit independently developed based on the SuperNAT technology platform, further strengthening the company's technological leadership and intellectual property barriers in the field of accurate diagnosis of kidney disease.
